A sense of dread hangs over Indignation , the first feature directed by James Schamus, former CEO of Focus Features. Schamus produced—and sometimes had a hand in writing—a litany of great films, working with Ang Lee on the likes of Crouching Tiger , Hidden Dragon and air jordan 6 history of jordan real vs fake diamonds wD8cnoxEgB
. His directorial debut, based on a 2008 Philip Roth novel, is exquisite to behold, carefully recreating its era (the action primarily takes place in 1951) and offering some pretty period costumes. But this is a bleak film, one whose undercurrent of morbidity stems any romanticization of the past. That ominousness can at times be suffocating, as the action barrels toward a conclusion it insists on foreshadowing. Light summer fare this is not.

The film’s more challenging and occasionally frustrating elements mirror those of its hero, Marcus Messner (Logan Lerman). Marcus, the son of a Jewish butcher in drearily lit Newark, New Jersey, has avoided being shipped off to the Korean War by getting a scholarship to a liberal arts college in Ohio. An early punchline comes when Marcus is asked by the grieving mother of a classmate killed in combat how he’ll keep kosher in the midwestern state. It quickly becomes clear that Marcus is not especially concerned with avoiding treif as he digs into some escargot on his first date with the striking Olivia Hutton (Sarah Gadon). A prickly figure who initially strives to keep to himself, Marcus can’t resist the starlet glamour of Olivia, and it’s through her that Marcus begins to shed his guarded armor, exposing a budding firebrand. He’s not a good Jewish boy trying to please his parents. He’s a fiercely independent atheist.

Olivia possesses a manic energy that instantly codes her as trouble. She’s got Donna Reed’s wardrobe and Tracy Flick’s cadence. Gadon is appropriately magnetic in this shiksa goddess role, but her performance is hampered by the nature of the part. The audience is only allowed to see Olivia through Marcus, and though she reveals to him that she attempted suicide in the past, he makes little effort to truly understand her, mostly regarding her as beautiful, intriguing, and very willing to service his penis. She gives him a blowjob on their first date, the significance of which he puzzles and agonizes over. Other acts follow. (Is this the hand job-iest movie of 2016? It is based on a Roth book.) While enjoying the pleasures she provides, Marcus refuses to acknowledge the full extent of Olivia’s pain, though she does admittedly conceal it from him. Thus, Gadon can’t betray much of the turmoil lurking under this young woman’s surface.

For those of us who work with Mus musculus or Homo sapiens , to name a couple of species, a few clicks on UCSC Genome Bioinformatics Site or Ensembl gets you the full and precise DNA sequence for any annotated gene in the genome. This luxury is not in place for all species however; many of which remain unsequenced.So how do we uncover the genetic code of our favourite model organism at our gene or region of interest?

Most of the time, help will come from other closely related species, for which the genetic sequence, or protein code of the gene we are interested in, is known. Homologous genes or amino acid sequences can be aligned from multiple species and conserved and non-conserved regions between species can be identified.

If we know the amino acid code of related organisms, we can work backwards to design a number of ‘degenerate’ primers of multiple iterations to find a set that are complementary to our ‘unknown’ sequence and give an amplifiable product.

A degenerate primer is defined as: “A mix of oligonucleotide sequences in which some positions contain a number of possible bases, giving a population of primers with similar sequences that cover all possible nucleotide combinations for a given protein sequence” (Iserte 2013) . For example:


refers to a series of primers in which the seventh and twelfth nucleotides are degenerate. The amount of degeneracy is defined by the number of different primer combinations in the mix. The above example has a degeneracy value of six .

Using the International Union of Pure and Applied Chemistry (IUPAC) system for degenerate bases (Table 1) we can insert a single-letter code to represent our unknown region. In the above example we would use ‘S’ in place of GC, and ‘V’ in place of AGC.

To design your own set of degenerate primers, follow some basic guidelines:

Depending on the goal, the play-off between primer specificity and efficiency can be modified by altering the degeneracy of the primer. For example, the more degenerate the primers, the less specific annealing will be; however, decreased degeneracy will allow more potential to identify unknown variants.

Thankfully, like basic primer design, there is help out there for designing degenerate primers. There are a number of online and downloadable programs available to aid design. Based on your input sequence, the software will generate the minimum number of degenerate primers while maintaining optimal PCR requirements. Some commonly used programs to try out include iCODEHOP (COnsensus-DEgenerate Hybrid Oligonucleotide Primers), NCBI Primer-BLAST or HYDEN (HighlY DEgeNerate primers) .

Basal vs. Prandial : Insulins can be divided into two categories based on function: basal (long-acting insulin) and prandial (rapid-acting or “mealtime” insulin). Basal insulin is designed to be injected once or twice daily to provide a constant level of insulin action throughout the day.
